The water content of adenosine disodium triphosphate was determined by Carsten moisture

In this paper, the water content of disodium adenosine triphosphate was determined by clamping method. Two kinds of reagents were used, one was to titrate with ψ(ethylene glycol: methanol)=60∶40 as solvent, and the other was to titrate with imported Carboldt reagent. The recoveries of the two reagents were 96.0% with RSD of 4.9%(n=10). 99.0%,RSD=315%(n=11). It is concluded that both domestic and imported Cartesian reagent can be used for the determination of water content of ADENosine disodium triphosphate and for the quality control of drugs.

Adenosine triphosphate (ATP) is a high energy compound existing in organisms, which directly participates in energy metabolism and can improve body metabolism as a coenzyme [1]. Medicinal ATP is disodium salt with three water of crystal [2]. The quality standard of ADENosine disodium triphosphate is not included in pharmacopoeia of various countries. Only specifications for Foreign Medicines (1997)[3] Ministry of Health drug Standards (1998)[4]. Its moisture determination in Japan using Kassler moisture residual titration method, the ministry standard using phosphorus pentoxide decompression drying method. In this paper, fischer titration method of adenosine disodium triphosphate was established by referring to the method of external prescription of Japanese pharmaceutical bureau. The method is simple, accurate and suitable for daily inspection and control of product quality.

1 Instrument and drug test

Metrohm 785 titration apparatus is equipped with type 703 cartesian water titration table, and ShimadzuLibrorAEU2130 electronic balance. Cartesian reagent (Tianjin Siyou KFR206 peerless pyridine Karl Fischer reagent), anhydrous methanol, ethylene glycol. German production (Sigma2Aldrich) Cartesian reagent (HydranalSolven, Hydranal2Titrant5) are commercially available.
